Primary DNA damage can be detected in the target tissue following chemical exposure. Gene mutations and/or DNA adducts or other measures of primary DNA

DNA adducts The initial step in chemical carcinogenesis is characterized by attachment of the chemical to DNA to produce DNA adducts. The chemicals can alter the structure of the DNA and in turn, the biological processing of the DNA by cellular proteins governing replication, transcription and repair. DNA adducts are typically present at trace levels in the human genome, requiring a very sensitive and comprehensive data acquisition and analysis method. Our software, wSIM-City, was created to process mass spectral data acquired by a wide selected ion monitoring (wSIM) with gas-phase fractionation and coupled to wide MS 2 fragmentation. DNA adducts that are exceptions to this paradigm are those (1) formed on the phosphate portion of DNA, (2) excreted as free nucleobases in urine because of spontaneous hydrolysis, depurination, and/or active repair, and (3) structurally similar to the DB[a,I]P dihydroepoxide adduct of dA, which upon fragmentation was found to produce mostly Methods: DNA from 32 intrahepatic CCa patients (and in 28 cases DNA from adjacent non-tumour tissue) and from biliary ducts of seven non-cancer patients were investigated for the presence of DNA adducts using the nuclease P1 method of 32P postlabelling. DNA adduct levels (number of adducts/108 nucleotides) were quantified.

In this work we report the attempt to detect this DNA-adduct using both an electrochemical assay Assignment on Genotoxicity 1.

The chemical can induce mutations, cytogenetic damage, DNA adducts and/or primary DNA damage in vitro. Modified from Moore (2008).

DNA adduct measurements provide valuable information about DNA damage associated with exposure to specific genotoxicants. N 2-Ethylidene- dGuo, the major DNA adduct formed upon reaction of acetaldehyde with DNA, has been used to investigate mechanisms of alcohol carcinogenesis focusing on the effects of acetaldehyde, the primary metabolite of ethanol.

12. Interindividual variation is observed in carcinogen metabolism and/or formation of carcino-gen-DNA adducts in human cells and tissues. 13. Normal cells … DNA adducts occur when a chemical covalently binds to DNA and can form in response to a chemical exposure, thus representing one type of DNA damage.

DNA adducts have been shown to inhibit DNA synthesis and the ability of adducts such as AF and AAF, to induce radically different effects during synthesis has been linked to the structures they adopt within the active site of polymerase and their ability to disrupt or block conformational changes.

Lindane DNA adducts are identified for the first time. The results link gene research with the nutritional and environmental approaches to medicine. Early indications are that the burden of DNA adducts may be reduced by nutritional intervention. rizing N2-dG adducts into three classes—small (up to 7 atoms), moderate (8–20 atoms), and bulky (> 20 atoms). A list of known N2-adducts along with the molecular formula and num-ber of atoms added is shown in Table 1.
